Single-Molecule Fluorophores for Cellular Imaging

چکیده
This P20 exploratory program brings together experts in synthetic chemistry, biophysics, single-molecule imaging in cells, and developmental microbiology to focus on three key areas: design and synthesis of new fluorophores for improved imaging in cells, photophysical assessment and utilization of new singlemolecule fluorophores in cellular targeting schemes, and application of new imaging methodologies to explore mechanisms of regulatory protein localization in Caulobacter crescentus.

Photophysical properties of acene DCDHF fluorophores: long-wavelength single-molecule emitters designed for cellular imaging.
We report the solvatochromic, viscosity-sensitive, and single-molecule photophysics of the fluorophores DCDHF-N-6 and DCDHF-A-6. These molecules are members of the dicyanomethylenedihydrofuran (DCDHF) class of single-molecule emitters that contain an amine electron donor and a DCDHF acceptor linked by a conjugated unit; DCDHF-N-6 and DCDHF-A-6 have naphthalene- and anthracene-conjugated linkers...
متن کاملThe covalent trimethoprim chemical tag facilitates single molecule imaging with organic fluorophores.
Chemical tags can be used to selectively label proteins with fluorophores that have high photon outputs. By permitting straightforward single molecule (SM) detection and imaging with organic fluorophores, chemical tags have the potential to advance SM imaging as a routine experimental tool for studying biological mechanism. However, there has been little characterization of the photophysical co...

متن کاملNovel fluorophores for single-molecule imaging.
Nonlinear optical chromophores based on dicyanodihydrofuran acceptors paired with amine donors have been found to exhibit sufficiently large fluorescence quantum yields and stability to enable single-molecule detection in polymeric hosts. To illustrate the breadth of this class, six fluorophores are presented, spanning the emission range from 505 to 646 nm. In contrast to conventional single-mo...

Bright, long-lasting organic fluorophores enable a broad range of imaging applications. "Self-healing" fluorophores, in which intra-molecularly linked protective agents quench photo-induced reactive species, exhibit both enhanced photostability and biological compatibility. However, the self-healing strategy has yet to achieve its predicted potential, particularly in the presence of ambient oxy...
